I want to make it a required field during sign up?

 

I am unable to see this field

Go to ACP -> Members -> Profile -> edit Gender filed. Enable "Required" then enable "Show on registration page"

I cannot see Gender field -  Do i need to create...  

Even DOB  is not visible

If this is a new install than yes, you will need to create this field. If it is an upgrade from 3.x and didn't remove it than it should have been upgraded.
