I have a single language forum and a single language file. I used the globe icon and translation tools in the ACP to tweak things like message content, error messages, labels, and to root out all references to Clubs and change them to Committee(s).

Will these survive our upgrade from v4 to v5? Or will I need to redo?

It will upgrade but you may encounter issues depending on what you changed in the language pack. Language strings will have changed between 4 and 5 so you will need to re-evaluate it.
